Cytochrome P450 2C9 (CYP2C9) is a monooxygenase anchored to the Endoplasmic reticulum membrane, its expression being in both hepatic and extrahepatic tissues. Hepatic CYP2C9 metabolizes drugs like warfarin and extraheapaendogenous compounds like Arachidonic acid, 5HT etc. CYP2C9 is implicated in clinically relevant drug/drug interactions. BioVision’s human CYP2C9 ELISA Kit is based on the standard principle of a sandwich enzyme-linked immunosorbent assay. A human CYP2C9-specific antibody is coated on the 96-well plate. Standards or samples are then added to the appropriate wells with a biotin-conjugated antibody specific to CYP2C9. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. After TMB substrate solution is added, only those wells that contain CYP2C9, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color to form a blue product. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change from blue to yellow is measured spectrophotometrically at 450nm . The density of yellow color is proportional to the human CYP2C9 captured onto the plate. The concentration of CYP2C9 in the samples is then determined by comparing the O.D. of the samples to the standard curve. This ELISA kit shows no detectable cross-reactivity with related isoforms.
