Cytochrome P450 1A2 (CYP1A2) is a monooxygenase anchored to the Endoplasmic reticulum membrane, its expression being highest in liver and intestinal and olfactory mucosal cells. CYP1A2 metabolizes ~10% of all drugs on the market. CYP1A2 is implicated in clinically relevant drug/drug interactions. BioVision’s human CYP1A2 ELISA Kit is based on the standard principle of a sandwich enzyme-linked immunosorbent assay. A human CYP1A2-specific antibody is coated on the 96-well plate. Standards or samples are then added to the appropriate wells with a biotin-conjugated antibody specific to CYP1A2. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. After TMB substrate solution is added, only those wells that contain CYP1A2, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color to form a blue product. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change from blue to yellow is measured spectrophotometrically at 450nm . The density of yellow color is proportional to the human CYP1A2 captured onto the plate. The concentration of CYP1A2 in the samples is then determined by comparing the O.D. of the samples to the standard curve. This ELISA kit shows no detectable cross-reactivity with related isoforms.
