Cytochrome P450 2D6 (CYP2D6) is a monooxygenase anchored to the Endoplasmic reticulum membrane, its expression being highest in liver and brain. CYP2D6 is responsible for metabolism of nearly 25% of all small molecule drugs commonly used by humans, particularly psychiatric drugs such as antidepressants, antipsychotics and stimulants. BioVision’s rat CYP2D6 ELISA Kit is based on the standard principle of a sandwich enzyme-linked immunosorbent assay. A CYP2D6-specific antibody is coated on the 96-well plate. Standards or samples are then added to the appropriate wells with a biotin-conjugated antibody specific to CYP2D6. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each well and incubated. After TMB substrate solution is added, only those wells that contain CYP2D6, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color to form a blue product. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change from blue to yellow is measured spectrophotometrically at 450nm . The density of yellow color is proportional to the rat CYP2D6 captured onto the plate. The concentration of CYP2D6 in the samples is then determined by comparing the O.D. of the samples to the standard curve. This ELISA kit shows no detectable cross-reactivity with related isoforms.
